Temporarily excluded whole blood donors at the collection center of the Hematology-Transfusion Department in Jihlava Hospital

This bachelor thesis is focused on the topic of Temporarily excluded whole blood donors at the collection center of the Hematology and Transfusion Department in the Jihlava Hospital. The theoretical part of the work generally deals with blood donation according to applicable regulations and recommendations. This section describes the criteria for donation, the course of blood donation, self-exclusion, the production of transfusion products, the donor registry, the valuation of blood donors and the risks associated with donation. The methodology of the work included measuring the blood count of donors. At the collection center in Jihlava, the result of the blood count is one of the donor's pre-collection examinations. The blood count of donors is measured on a Sysmex XN 1000 analyzer. The values of hemoglobin, leukocytes and platelets are especially important. The main goal of the bachelor thesis was to find out the reasons for the temporary exclusion of whole blood donors at the collection center in Jihlava. The practical part was developed on the basis of data from the information systems HEMO and TIS. The reasons for the temporary exclusion were evaluated for the period 2000-2020. Another goal of the work was to find out how donors are informed at the collection center. Part of the practical part was a questionnaire for blood donors, which served to verify the most common reasons for temporary exclusion. The second part of the questionnaire was about the awareness and satisfaction of donors at the collection center. According to the results of the work, donors are most often temporarily excluded due to low levels of hemoglobin (red dye). It is also obvious that women are more often excluded than men. According to the donors, the awareness at the collection center in Jihlava is very good, which follows from the results of the questionnaire. In some cases, the increased awareness can contribute to reducing the number of temporarily rejected blood donors.
Issue of diagnosis and treatment of sideropenic anemia

Iron deficiency anemia is the most common type of anemia. It occurs in all countries of the world and it is mostly dependent on the standard of living. It is characterized by iron deficiency, which results in a reduced quality of life for adults, impaired child growth and the risk of abortion for pregnant women. Iron deficiency anemia can be diagnosed by hematological and biochemical blood tests. Iron deficiency is usually a secondary disease so the treatment of iron deficiency also depends on the treatment of primary desease. The practical part focuses on the topic of blood count and the examination of the feritin level with the automatic analysers. These parameters were defined for 15 anaemic patients with that were treated at the haematology department and at the clinical biochemistry department of the Hospital in Jindrichuv Hradec. The goal of the thesis is to determine the iron and feritin levels blood count for those patients and consequently establish a connection between the levels and the treatment of the anemia.
Analysis of the impact of a 10-day training camp at a higher altitude on performance of adolescent runners

Goal of this Bachelor thesis is to check, if there will be any changes of functional and biochemical parameters of the organism for medium and long length track adolescent runners during a 10-day training camp in high altitude around 1850 meters. Runners were subjected to functional laboratory examination both before and after the camp. All requisites were identical for both the control group and the runners who completed this camp. We were examining measured parameters of VO2max, resting heart rate, breath capacity, expiratory minute ventilation, pulse oxygen, respiratory rate, respiratory exchange ratio, heart rate during peak oxygen consumption and maximum watt performance. Most significant watched parameter was VO2max, where we saw an increase of 15,52 % from original 55,40 +- 7,73 ml-min-1-kg-1 to 64,00 +- 9,87 ml-min-1-kg-1 . Another one was resting heart rate, there was a decrease of 5,86 % for the runners after coming back to lowland compared to rates that were measured before the camp. Erythrocytes values increased by 2,38 %, haemoglobin by 2,69 % and haematocrit by 2,27 %. All these results show factual and statistical importance, the only parameter that didn't show any statistical importance was haematocrit. Result of our research confirmed, that 10-day training camp has a major impact on functional and biochemical parameters of the organism.

The thesis is focused on the comparison of the influence of different types of starters on feed intake, holstein breed calf increments, feed conversion, economy, rumen fluid and blood parameters during calf rearing. After the colostrum period, the calves were divided into 3 groups of 5 individuals, they were fed by dairy feed mixture and various types of starter mixtures. The first group had ČOT – B granular starter (granulated granulated components), the second group received Premiumstart – muesli and the third group got Goldstart – a mixture of granules, corn flakes and cut straw ("straw starter"). In 60 days old they were bled and then killed in a slaughterhouse. After slaughter, the rumen and rumen fluid were collected from the calves for subsequent analyzes. The average starter consumption and feed conversion was the highest in the second group. The average daily increase was relatively balanced in the individual groups. The load per kg increment and the real starter consumed was demonstrably the lowest in the first group. The average pH of rumen fluid was significantly lower in the first group (below 5.5). Propionic acid was relatively high in all calves (up to 15% in the first group, 9% in the second and third groups) and low levels of acetic acid (8% lower). A low number of ciliates was found in all groups, but in the third group level was close to the reference values of adult cattle (119 thousand / ml). In calves received Goldstart starter straw, rumen fermentation values were closest to the physiological rumen environment. In this group of pH, the number of ciliates and the proportion of acetic acid and propionic acid corresponded. In terms of rumen papillum development, rumen volume, ammonia, pH, WBC, and ALP levels, Premiumstart – muesli is the most appropriate. The WBC concentration was highest in calves that received muesli starter. The highest glucose levels were observed in calves that received the ČOT – B granulator starter. The highest levels of ALP were found in the calves that received the muesli starter, which can be confirmed by the highest daily gains in these calves. The results obtained give a good idea of the functioning of the individual starter mixtures. However, for practice, it will be necessary to repeat the experiment to confirm some claims.
Comparison of values of blood images in animals - dog, cat

Blood count is one of the most important and most frequent haematological examinations. It is important for proper diagnosis and control of treatment effectiveness. Non-coagulated blood with the anticoagulant K3EDTA or K2EDTA (ethylenediaminetetraacetic acid potassium salt) is used to examine the blood count.It is also important to maintain the correct ratio of anticoagulant and blood collected. Examination of blood counts is not related only to human patients, but also to many species of animals, most often dogs and cats. In order to evaluate the results correctly, it is necessary to know the reference limits of the respective species, possibly divided by sex, age or breed. The aim of this study is to evaluate differences in baseline blood counts (amount of erythrocyte, amount of hemoglobin, value of hematocrit, amount of leukocyte, amount of platelet) between samples obtained from dogs and from cats. A total of 500 samples (250 canine, 250 cat) was measured on a Sysmex XN 1000 analyzer in Synlab czech, s.r.o., Ceske Budejovice according to proper laboratory practice procedures. Animal patients are selected regardless of diagnosis, breed and sex. The measured results are evaluated by the parametric method, the correlation coefficient is always calculated. This correlation coefficient was close to 1 for all five monitored parameters. This means that the individual parameters of dogs and cats are comparable between species regardless of age. Results of dogs and cats in most cases exceed the values of reference limits given to a specific species of animal. It can be assumed that the reason for the high values is that samples of diseased animals were predominantly used.
Cold antibody capture in a hematology laboratory while performing a blood count.

In my Bachelor thesis I followed up cold antibodies capture in a hematology laboratory during the blood cell count statement. The main goal of this thesis was to point out the possible influence of blood counts through the cold antibodies, practical laboratory methods - measurement of blood count values with hematological Abbott analyser, furthermore influencing the results of setting the cold antibodies through preanalytical phase and finally the assessment of received results in amount of 20 patients. The first part of this thesis focuses on general information of imunohematology which describes antigen, antibodies, structure of immunoglobulins. Moreover I dealt with physiology and blood composition, the blood counts and its parameters. In the end of this thesis there are information of autoimmune hemolytic anemia and cold antibodies including their preanalytical, analytical and postanalytical phase. The methodological part of this thesis was carried out in Biochemical-hematological laboratory Stafila s.r.o. All my measurements were made under the supervisory of hematological laboratory assistant. Stating the blood count was made with 24 patients through multiparametric fully automatic Abbott analyser. In that part, I firstly describe analyser specifications. In the following step I indicate the working aids which were used. Finally, there is described the process of the measurements in detail. Within the assessment of the measured values the main attention was paid to red blood line, especially influencing the values of erythrocyte, the hemoglobin, MCH and MCHC. In another part of this thesis are all the blood counts calibres of 24 samples, which I entered into graphs and charts. Out of all 24 measured values I can evaluate that only 4 samples were of unusual values of red line, which can indicate the presence of cold antibodies.
Monitoring of blood counts parameters during the therapy in patients with myelodysplastic syndrome

Blood counts belongs among basic examination methods in hematology laboratory. It is used to determine the basic parameters of leukocytes, erythrocytes, hemoglobin, hematocrit and platelets. The goal of study was to determine which blood counts have changed during the treatment of patients with myelodysplastic syndrome using immunomodulatory drug lenalidomide. Analysis of blood samples were performed on the Sysmex XE-5000. The selected blood counts parameters were evaluated in two periods - before the beginning of the treatment and during the treatment. The final blood counts showed increasing concentrations of hemoglobin and decreasing platelet values; other blood count parameters have not changed significantly. The evaluated blood counts changes could be used to determine the efficiency of the treatment and the transfusion therapy could be stopped based on these results.
